Project title: An expedient route to chiral spirocyclic frameworks

Summary: Spirocyclic scaffolds are considered ‘privileged structures’ that provide an exciting platform for the development of novel three-dimensional structures in drug discovery. Indeed, the specific spatial arrangement of these compounds leads to a semi-rigid molecular framework that offers conformational restriction, thereby, reducing the entropic penalty when bound to the receptor. Nonetheless, spirocyclic derivatives still remain underexploited, which can be attributed to the lack of general methods that furnish the key quaternary stereogenic centre in a robust, efficient, and highly stereoselective fashion. The aim of this research project is to develop simple, scalable and highly versatile methods for the synthesis of chiral spirocyclic compounds with complete control of connectivity and stereochemistry.
